The Walsh lab uses both experimental and computational approaches to understand the genetic basis of cancer. Lung cancer is responsible for over 25% of all cancer related deaths. One in twelve Canadians will develop lung cancer during his/her lifetime and more people die from lung cancer than breast, colorectal, and prostate cancer combined. The Walsh lab is focused on translational research questions, as we foster close collaborations between physicians and scientists to address clinically relevant challenges.

- Biomarqueurs L'affectation des patients atteints de cancer du poumon à la variété des thérapies conventionnelles et nouvelles disponibles, telles que l'immunothérapie, est actuellement informée par des données très rudimentaires. En conséquence, prédire les taux de réponse aux traitements systémiques conventionnels, à l'immunothérapie mono ou multi-cible, ou aux approches combinatoires est difficile et peut entraîner des résultats sous-optimaux pour les patients, tant en termes de réponse que de toxicité. Biomarkers Assignment of patients with lung cancer to the variety of available conventional and novel therapeutics such as immunotherapy is currently informed by highly rudimentary data. As a result, predicting response rates to conventional systemic therapy, single or multi-target immunotherapy or combinatorial approaches is difficult and may lead to sub-optimal patient outcomes both regarding response and toxicity. The lab uses AI combined with next-generation sequencing technology to define how and when, amongst the available armamentarium of systemic therapies, which are optimally utilized by delineating their mechanisms and the contexts within which they perform best.
- Métastase Malgré la métastase étant la principale cause de décès lié au cancer, nous manquons d'une compréhension unifiée de la reprogrammation phénotypique à grande échelle sous-jacente à la maladie métastatique. Alors que plusieurs gènes suffisants pour induire la progression métastatique ont été identifiés, il existe une valeur inexploitée dans l'identification systématique de gènes qui sont plus universellement nécessaires pour la progression, en tant que cibles thérapeutiques précieuses. Le laboratoire Walsh interroge les réseaux d'interaction transcriptionnelle spécifiques au cancer pour élucider les structures de contrôle transcriptionnel causalement responsables de la régulation des programmes génétiques activés lors de la transition de la tumeur primaire vers les métastases. Metastasis Despite metastasis being the leading cause of cancer‐related death, we lack a unifying understanding of the large‐scale phenotypic reprogramming underlying metastatic disease. While several genes that are sufficient to induce metastatic progression have been identified, there is untapped value in the systematic identification of genes that are more universally necessary for progression, as valuable therapeutic targets. The Walsh lab interrogates cancer specific transcriptional interaction networks to elucidate the transcriptional control structures causally responsible for regulating genetic programs activated during primary tumor transition to metastases.
- Technologies spatiales des cellules individuelles Les technologies à cellule unique ont apporté un changement transformateur dans notre compréhension de la biologie du cancer. Ces méthodes avancées nous permettent d'analyser les profils génomiques, transcriptomiques et protéomiques des cellules individuelles au sein d'une tumeur, fournissant un niveau de détail qui était auparavant inatteignable. Cette vision granulaire est cruciale car le cancer n'est pas une maladie uniforme ; c'est un mélange complexe et hétérogène de différents types de cellules, chacun contribuant potentiellement différemment à la progression de la maladie et à la réponse au traitement.
De plus, la résolution spatiale est un aspect essentiel de cette nouvelle ère de la recherche sur le cancer. Il ne s'agit pas seulement de savoir quelles cellules sont présentes dans une tumeur, mais aussi de comprendre où elles se trouvent et comment elles interagissent les unes avec les autres. Les technologies spatiales nous permettent de cartographier les emplacements des différents types de cellules au sein du microenvironnement de la tumeur. Ce contexte spatial ajoute une couche d'information inestimable, révélant comment l'arrangement et l'interaction des cellules influent sur le comportement de la tumeur et les résultats pour les patients.
Notre laboratoire développe et affine des outils qui interrogent le microenvironnement immunitaire de la tumeur avec une résolution spatiale. En examinant comment les cellules immunitaires et les cellules cancéreuses interagissent in situ, nous visons à découvrir de nouvelles perspectives sur les mécanismes d'évasion et de suppression immunitaires utilisés par les tumeurs. Cela peut conduire à l'identification de nouvelles cibles thérapeutiques et au développement de traitements contre le cancer plus efficaces et personnalisés. Notre focus sur les technologies à résolution spatiale représente une étape cruciale dans l'avancement de notre compréhension de l'interaction complexe entre les cellules cancéreuses et le système immunitaire au sein du paysage tumoral.
Single cell spatial technologies Single cell technologies have brought about a transformative change in our understanding of cancer biology. These advanced methods allow us to analyze the genomic, transcriptomic, and proteomic profiles of individual cells within a tumor, providing a level of detail that was previously unattainable. This granular insight is crucial because cancer is not a uniform disease; it's a complex, heterogeneous mix of various cell types, each potentially contributing differently to the disease's progression and response to treatment.
Furthermore, spatial resolution is a critical aspect of this new era of cancer research. It's not just about knowing which cells are present in a tumor, but also understanding where they are located and how they interact with each other. Spatial technologies enable us to map the locations of different cell types within the tumor microenvironment. This spatial context adds an invaluable layer of information, revealing how the arrangement and interaction of cells influence tumor behavior and patient outcomes.
Our lab develops and refines tools that interrogate the tumor immune microenvironment with spatial resolution. By examining how immune cells and cancer cells interact in situ, we aim to uncover novel insights into the mechanisms of immune evasion and suppression employed by tumors. This can lead to the identification of new therapeutic targets and the development of more effective, personalized cancer treatments. Our focus on spatially resolved technologies represents a crucial step in advancing our understanding of the complex interplay between cancer cells and the immune system within the tumor landscape.
